摘要:
目的探讨外周血单个核细胞(peripheral blood mononuclear cell,PBMNC)中核糖体蛋白(ribosonoal protein S 26,RPS 26)和卵巢颗粒细胞中过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptor gamma,PPARγ)基因表达与多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者病情的相关性。方法将深圳市龙华新区人民医院妇科2015年7月至2016年3月诊治的38例PCOS患者作为PCOS组,同期体检健康女性30例作为对照组,抽取外周血,采用Ct值比较法,通过SYBR Green II 实时定量PCR检测两组PBMNC中RPS 26及PPARγ基因在mRNA的表达水平,进一步分析PCOS患者两基因表达水平及与体质量指数(body mass index,BMI)、性激素水平相关性。结果PCOS组RPS 26表达水平高于对照组,但差异无统计学意义(P>005),PCOS组PPARγ基因表达明显高于对照组,差异有统计学意义(P<005)。RPS 26及PPARγ基因表达水平与睾酮呈正相关(P<005),与促卵泡成熟素呈负相关(P<005)。结论RPS 26及PPARγ基因在PCOS患者PBMNC中的表达均高于正常女性,可不同程度地反映疾病的严重程度。
Abstract:
ObjectiveTo investigate correlation of the expression of RPS 26 and PPARγ in peripheral blood mononuclear cells and condition of patients with PCOS. Methods38 cases of PCOS patients and 30 cases of healthy women as control were collected in People’s Hospital of Longhua New District from July 2015 to March 2016. Peripheral blood was drawn and detected with CT value comparison method. RPS 26 and PPARγ gene expression in peripheral blood single mononuclear cells (PBMNC) level was analyzed by SYBR Green II real-time quantitative PCR with further analysis of correlation in gene expression levels of PCOS patients,their body mass index (BMI)and gonadal hormone level. ResultsThe expression level of RPS 26 in PCOS group was higher than that in control group, but with no statistical difference (P>005). The expression of PPARγ in PCOS group was significantly higher than that in control group, with statistical difference (P<005). RPS 26 and PPARγ gamma gene expression were significantly positively correlated (P<005). RPS 26 and PPARγ gene expression levels were positively correlated with testosterone(P<005), and negatively correlated with follicle-stimulating hormone(FSH) (P<005). ConclusionThe expression of RPS 26 and PPAR gene in PBMNC of patients with PCOS are higher than those in normal women, and correlated with the severity of disease.
