摘要:
目的探究microRNA-206(miRNA-206)和胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)在早发型子痫前期(early onset preeclampsia,EOPE)孕妇血清中的表达水平及临床意义。方法选择2016年1月至2017年2月广东省妇幼保健院收治的105例孕妇为研究对象,包括35例正常晚期妊娠孕妇(正常妊娠组)、35例轻度EOPE孕妇(轻度EOPE组)和35例重度EOPE孕妇(重度EOPE组)。所有孕妇均在入院次日清晨空腹状态下采集肘静脉血,实时定量PCR(qRT-PCR)技术检测各组血清miRNA-206水平,酶联免疫(ELISA)检测各组血清IGF-1水平;利用受试者工作特征(ROC)曲线分析血清miRNA-206与IGF-1水平对EOPE的诊断效能。结果① 轻度、重度EOPE组血清miRNA-206水平均高于正常妊娠组(t=2689、5404,P=0009、0000),重度EOPE组高于轻度EOPE组(t=2139,P=0036)。② 轻度、重度EOPE组血清IGF-1水平均低于正常妊娠组(t=2853、5136,P=0006、0000),重度EOPE组低于轻度EOPE组(t=2201,P=0031)。③ EOPE患者血清miRNA-206水平与IGF-1水平呈负相关关系(r=-0364,P=0002)。④ ROC曲线分析结果发现,血清miRNA-206诊断EOPE的曲线下面积为0749,其诊断的敏感度为6768 %、特异度为7372 %;血清IGF-1诊断EOPE的曲线下面积为0668,其诊断的敏感度为4002 %、特异度为9677 %。结论EOPE患者miRNA-206表达水平较健康孕妇高、IGF-1血清表达水平较健康孕妇低,提示监测血清miRNA-206、IGF-1水平对早期诊断EOPE具有一定的参考价值。
Abstract:
ObjectiveTo explore the expression levels and clinical significances of microRNA-206 (miRNA-206) and insulin-like growth factor-1 (IGF-1) in the serum of pregnant women with early onset preeclampsia (EOPE).MethodsA total of 105 pregnant women in Maternal and Child Health Hospital of Guangdong Province from January 2016 to February 2017 were selected, including 35 cases of normal late pregnant women, 35 cases of mild EOPE pregnant women and 35 cases of severe EOPE pregnant women. All pregnant women were collected the venous blood in the morning of the next day of admission to the hospital, and measured the level of miRNA-206 in each group by real-time quantitative PCR (qRT-PCR); the serum levels of IGF-1 were detected by enzyme linked immunosorbent assay (ELISA); the diagnostic efficacy of serum miRNA-206 and IGF-1 levels on EOPE was analyzed by the ROC curve. Results① The level of serum miRNA-206 in the mild EOPE group and severe EOPE group was higher than that in the normal late pregnancy group (t=2689、5404, P=0009、0000), the severe EOPE group was higher than that the mild EOPE group (t=2139, P=0036).② The level of serum IGF-1 in the mild EOPE group and severe EOPE group was lower than that in the normal late pregnancy group (t=2853、5136, P=0006、0000),the level of serum IGF-1 in the severe EOPE group was lower than that in the mild EOPE group (t=2201, P=0031).③ The level of serum miRNA-206 was negatively correlated with the level of IGF-1 in patients with EOPE (r=-0364, P=0002).④ The results of ROC curve analysis showed that, the area under the curve of serum miRNA-206 for diagnosis of EOPE was 0749, the sensitivity of the diagnosis was 6768 %, and the specificity was 7372 %; the area under the curve of serum IGF-1 for diagnosis of EOPE was 0668, and the sensitivity of the diagnosis was 4002 %, and the specificity was 9677 %. ConclusionThe expression level of miRNA-206 was higher than that of healthy pregnant women and IGF-1 lower than that of healthy pregnant women, it is suggested that the monitoring of serum miRNA-206 and IGF-1 levels has a certain reference value for early diagnosis of EOPE.
