摘要:
目的探讨人γ干扰素诱导蛋白16(interferon-inducible protein 16 ,IFI 16)在子痫前期孕妇胎盘组织中表达水平及其与滋养细胞侵袭和胎盘缺氧的相关性。方法选取中国医科大学附属盛京医院2016年11月至2017年12月收治的106例子痫前期孕妇作为观察组,同期正常健康孕妇106例为对照组,比较两组孕妇胎盘组织中IFI 16、IFI 16 mRNA的表达水平。结果观察组24 h尿蛋白定量值、舒张压、收缩压高于对照组,总蛋白、白蛋白水平低于对照组; IFI 16 mRNA及IFI 16的相对表达水平均高于对照组,上述各项两组比较差异均有统计学意义(P<005);观察组孕妇胎盘组织中IFI 16表达量中位数为100,据此将观察组孕妇分为高IFI 16组(56例)和低IFI 16组(50例),高IFI 16组孕妇基质金属蛋白酶-9(matrix metalloprotein-9,MMP-9)、血管内皮生长因子(vascular endothelial growth factor,VEGF)、胎盘生长因子(placental growth factor,PLGF) 水平低于低IFI 16组及对照组,可溶性fms-样酪氨酸激酶受体-1(soluble fms-like tyrosine kinase receptor-1,sFlt 1)、缺氧诱导因子-1α 、缺氧诱导因子脯氨酸羟化-1、瘦素表达水平高于低IFI 16组及对照组,差异均有统计学意义(P<005)。结论相对正常健康孕妇,IFI 16在子痫前期孕妇胎盘组织中表达水平较高,随IFI 16表达水平的上升,滋养细胞侵袭,MMP-9、VEGF、PLGF水平下降;胎盘缺氧,缺氧诱导因子-1α 、缺氧诱导因子脯氨酸羟化-1、瘦素水平反之升高。
Abstract:
ObjectiveTo investigate the expression of interferon-inducible protein 16(IFI 16) protein in placental tissue of preeclampsia and its correlation with trophoblast invasion and placental anoxia. Methods106 cases with preeclampsia in Shengjing Hospital Affiliated to China Medical University from November 2016 to December 2017 were selected as the observation group,another 106 healthy pregnant women in the same period were selected as control group. The expression of IFI16 protein and IFI16 mRNA in the placental tissue of two groups were compared. ResultsThe 24h urine protein value, diastolic blood pressure, systolic blood pressure of the observation group were significantly higher than those in the control group,and total protein and albumin levels were lower than the control group; the relative expression levels of IFI 16 mRNA and IFI 16 were higher in the observation group than those of the control group (P<005). The median of relative expression of IFI 16 in observation group was 1.00, based on this, the observation group was further divided into high IFI 16 group (56 cases) and low IFI 16 group (50 cases); the high IFI 16 group had lower levels of matrix metalloprotein-9 (MMP-9), vascular endothelial growth factor (VEGF), and placental growth factor (PLGF), as well as higher expression levels of soluble fms-like tyrosine kinase receptor-1 (sFlt1), hypoxia inducible factor(HIF)-1α, hypoxia-inducible factor proline hydroxylation-1(HPH-1), leptin than those of the low IFI 16 group and control group (P<005). ConclusionCompared with normal healthy pregnant women, the expression level of IFI 16 in placenta tissue is higher in preeclampsia placenta. With the increase of IFI16 expression level, the level of MMP-9, VEGF and PLGF in trophoblastic cells decrease, and the level of HIF-1α,HPH-1 and leptin increase.
