摘要:
目的检测子宫内膜异位症(以下简称内异症)患者异位、在位内膜组织中干扰素诱导蛋白1(interferoninduced protein with tetratricopetide repeats 1,IFIT 1)、干扰素诱导跨膜蛋白3(interferoninduced transmembrane protein 3,IFITM 3)表达,并探究其对子宫内膜间质细胞的增殖作用。方法选取2018年3月至2019年10月于武汉市第一医院就诊并行腹腔镜或开腹手术治疗、术后经病理证实为内异症的患者86例为研究对象,选取其在位内膜及异位内膜组织分别为在位内膜组与异位内膜组;选取同期因子宫肌瘤或宫颈病变行子宫切除术、病理证实为正常子宫内膜的患者74例为对照组。采用实时荧光定量PCR(qRTPCR)法检测组织中IFIT 1 mRNA、IFITM 3 mRNA表达水平,免疫组织化学法检测各组织中IFIT 1、IFITM 3蛋白表达,采用CCK8法检测IFIT 1、IFITM 3在子宫内膜异位间质细胞分裂增殖中的作用;分析内异症患者在位内膜组织、异位内膜组织中IFIT 1 mRNA、IFITM 3 mRNA表达水平相关性。结果内异症异位内膜组中IFIT 1 mRNA、IFITM 3 mRNA表达水平及其蛋白阳性表达率高于内异症在位内膜组和对照组,内异症在位内膜组中IFIT 1 mRNA、IFITM 3 mRNA表达水平及其蛋白阳性表达率高于对照组,差异均有统计学意义(P<005);内异症患者在位内膜组织、异位内膜组织中IFIT 1 mRNA、IFITM 3 mRNA表达水平均呈正相关(r=0534、0574,P<005);IFIT 1、IFITM 3可促进子宫内膜间质细胞的增殖和分裂,该作用随着IFIT 1、IFITM 3剂量的增加而增强;当IFIT 1、IFITM 3剂量增加到10 μg/L时,其促进子宫内膜间质细胞的增殖和分裂作用是对照组的186和188倍,差异有统计学意义(P<005)。结论IFIT 1、IFITM 3在内异症异位内膜组织、在位内膜组织中表达明显高于正常子宫内膜组织,且对子宫内膜间质细胞的增殖和分裂有促进作用。
Abstract:
ObjectiveTo detect the expressions of interferoninduced protein with tetratricopetide repeats 1 (IFIT 1) and interferoninduced transmembrane protein 3 (IFITM 3) in ectopic and eutopic endometrium of endometriosis patients, and to explore their effect on the proliferation of endometrial stromal cells.MethodsFrom March 2018 to October 2019,86 patients with endometriosis who were treated by laparoscopy or laparotomy in Wuhan NO.1 Hospital and confirmed by pathology after operation were selected as the study objects, the eutopic endometrium and ectopic endometrium were selected as in eutopic endometrium group and ectopic endometrium group respectively; at the same time, 74 cases of normal endometrium who were treated by hysterectomy due to hysteromyoma or cervical lesions and confirmed by pathology after operation were selected as the control group. The expression levels of IFIT 1 mRNA and IFITM 3 mRNA were detected by realtime fluorescent quantitative PCR (qRTPCR), the protein expressions of IFIT 1 and IFITM 3 was detected by immunohistochemistry, the effects of IFIT 1 and IFITM 3 on the proliferation of endometrial stromal cells were detected by CCK8 method; and the correlation between the expression levels of IFIT 1 mRNA and IFITM 3 mRNA in eutopic and ectopic endometrium of patients with endometriosis was analyzed.ResultsThe expression levels and protein positive rates of IFIT 1 mRNA and IFITM 3 mRNA in ectopic endometrium group were significantly higher than those in eutopic endometrium group and normal control group (P<005), the expression levels and protein positive rates of IFIT 1 mRNA and IFITM 3 mRNA in eutopic endometrium group were significantly higher than those in normal control group (P<005); there was a positive correlation between the expressions of IFIT 1 mRNA and IFITM 3 mRNA in eutopic and ectopic endometrium tissues (r=0534,0574,P<005);IFIT 1 and IFITM 3 can promote the proliferation and division of endometrial stromal cells, and the effect was enhanced with the increases of IFIT 1 and IFITM 3; when the doses of IFIT 1 and IFITM 3 increased to 10 μg/L, their effects of promoting the proliferation and division of endometrial stromal cells were 186 and 188 times higher than those of the control group (P<005).ConclusionThe expressions of IFIT 1 and IFITM 3 in ectopic endometrium tissues and eutopic endometrium tissues of endometriosis are significantly higher than those in normal endometrial tissue, and they can promote the proliferation and division of endometrial stromal cells.
