摘要:
目的检测不同月经周期子宫内膜息肉(endometrial polyps,EP)及正常子宫内膜组织中半胱氨酸天冬氨酸蛋白酶(Caspase)-8和Caspase-3的表达水平,探讨Caspase凋亡通路在EP发生发展中的作用。方法选取2011年12月至2012年5月在哈尔滨医科大学附属第一医院就诊的120例患者,采用反转录酶-聚合酶链反应法和免疫组化法分别检测正常子宫内膜组织56例(对照组)及EP组织 64例(息肉组)不同月经周期中Caspase-8和Caspase-3基因和蛋白的表达水平。 结果Caspase-3在两组腺上皮细胞的胞浆、胞膜上均有表达,Caspase-8主要表达于两组腺上皮细胞胞浆中;增生期两组标本中Caspase-8及Caspase-3均呈弱表达,差异无统计学意义(P>005);分泌期息肉组Caspase-8及Caspase-3的表达水平均高于增生期 (P<005),Caspase-8及Caspase-3在息肉组的表达均显著低于对照组(P<005)。结论Caspase-8和Caspase-3均参与了子宫内膜由增生期向分泌期的转变,Caspase-8和Caspase-3在分泌期EP中表达低于增生期,提示Caspase通路可能参与EP的发生发展。
Abstract:
ObjectiveTo detect the expression of Caspase-8 and Caspase-3 in endometrial polyps(EP) and normal endometrial tissue in different menstrual cycle, and to investigate the significance of the apoptotic Caspase pathways in the occurrence and development of endometrial polyps. Methods120 patients hospitalized in the First Affiliated Hospital of Harbin Medical University from December 2011 to May 2012 were selected. Detected the expressions of mRNA and protein of Caspase-8 and Caspase-3 in normal endometrium of 56 patients(the control group) and EP of 64 patients(the EP group) in different menstrual cycle by immunohistochemistry and reverse transcription-polymerase chain reaction(RT-PCR). ResultsCaspase-8 was mainly expressed in normal endometrium and the cytoplasm of glandular epithelial cells of polyps. Caspase-3 was expressed in normal endometrium and cytoplasm and cell membrane of glandular epithelial cells of polyps. There were no significant differences in the levels of Caspase-8 and Caspase-3 between the EP group and the control group in proliferative phase. Both were weakly expressed(P>005). The expression of Caspase-3 and Caspase-8 in the EP group in secretory phase was higher than that in poliferative phase(P<005). The expression of Caspase-3 and Caspase-8 in the EP group was significantly lower than that in the control group(P<005). ConclusionBoth Caspase-3 and Caspase-8 are involved in the transition from the proliferative phase to the secretory phase.The expression of Caspase-3 and Caspase-8 in endometrial polyps is decreased in secretory phase compared to proliferative phase, which proves that the Caspase signalings play an important role in the pathogenesis and development of endometrial polyps.
