摘要:
目的检测胚胎培养液中转化生长因子-β1(tansforming growth factor,TGF-β1)及血管内皮生长因子(vascular endothelial growth factor,VEGF)的浓度,分析二者的表达与胚胎质量及临床妊娠率的关系。方法收集唐山市妇幼保健院2017年3~12月接受不孕治疗43例患者Day 3胚胎培养液,采用酶联免疫吸附试验检测不同形态学评级胚胎培养液中TGF-β1、VEGF的表达;留取移植后培养液,检测不同妊娠结局各因子表达情况。免疫印迹法检测每组样本中各因子蛋白浓度,比较各因子表达与胚胎评分的相关性。结果TGF-β1在Ⅰ、Ⅱ级胚胎培养液中的表达显著高于Ⅲ、Ⅳ级胚胎,在妊娠组中表达高于未妊娠组;而VEGF在Ⅰ级、Ⅱ级胚胎培养液中浓度明显低于Ⅲ、Ⅳ级胚胎,在妊娠组中表达降低,差异均有统计学意义(P<005)。TGF-β1与胚胎形态学评分呈正相关,VEGF与胚胎评分呈负相关。结论检测培养液中TGF-β1、 VEGF浓度或许可为评估胚胎质量及预测胚胎发育潜能提供新的理论依据。
Abstract:
Objective To detect the concentrations of transforming growth factor-β1 (TGF-β1) and vascular endothelial growth factor (VEGF) in embryo culture medium and investigated the relationship between the expression of the growth factors and embryo quality and pregnancy rate. Methods The culture medium of usable embryo was collected on the third day from patients receiving infertility treatment in Tangshan Maternity and Child Health Care Hospital from March to December in 2017. The concentrations of TGF-β1 and VEGF were measured by ELISA and the expressions of TGF-β1 and VEGF protein were evaluated by western blot. The correlation between the expression of each growth factor and embryo morphology score in each group was compared. ResultsCompared with the grade Ⅲ and grade Ⅳ groups, the expression of TGF-β1 in good quality embryo culture medium on the level of both concentrations detection and protein expression increased significantly. While VEGF decreased significantly in gradeⅠand grade Ⅱ groups. And the expression of TGF-β1 in pregnancy group was higher than that of non- pregnancy group oppositing to VEGF. The differences were statistically significant(P<005). TGF-β1 was positively correlated with embryo morphology score, while VEGF was negatively correlated with embryo score. ConclusionThe expression level of TGF-β1 and VEGF in embryo culture medium can be used for evaluating the embryo quality and predicting embryo developmental potential.
